    The study aims to examine the levels of cytokines in the serum of individuals with type 1 diabetes mellitus and compare these levels to those of healthy controls. This research aimed to assess the levels of some cytokines that includes IL-4, IL-9, IL-13, IL-35, IL-12, IL-17, IL-21 and IL-33 in Type I diabetes mellitus. In this study, 60 samples were collected from patients and 30 samples from healthy individuals, collected during the period from the of July 2023 to the March 2024.Results of this study showed a significant increase in the concentration of IL-4, IL-9, IL-13, IL-35, IL-12, IL-17, IL-21 and IL-33 in the serum of type 1 diabetes mellitus with the control group there were [10.22±2.065, 73.983±5.117, 35.90±3.343, 3.10±1.612, 19.80±3.433, 20.86±4.010,70.75±8.57, and 398.71±47.157, respectively] than in healthy groups [6.793±0.924, 47.00±5.535, 20.72±2.534, 1.01±10.597, 10.34±2.036, 10.79±1.818, 44.83±7.201 and 277.59±39.451, respectively. The results of the current study showed an imbalance in cytokines production that includes IL 4, IL 9, IL 13, IL 35, IL 12, IL 17, IL 21 and IL 33 in type 1 diabetes mellitus [T1DM] patients. The imbalance between pro- and anti-inflammatory cytokines could contribute to pancreatic β-cell destruction, offering insights for potential biomarkers and future therapies.     L-asparaginase is an amidohydrolase enzyme that is widely identified as one of the most potential anti cancerous drugs. Nevertheless, this drug is poorly bioavailable and hence its pharmaceutical uses are limited. To improve its bioactivity, L-asparaginase was loaded on gold nanoparticles (GNPs) along with Arg-Gly-Asp (RGD) peptide that direct the enzyme to targeted cancer cells with aim to enhancing the anticancer efficiency. Successful preparation of GNPs and the conjugate (GNPs-PEG-L-asparaginase-RGD) was verified and delineated by employing UVeVIS spectrophotometer, FTIR, XRD, FE-SEM, and TEM. UVeVIS spectrophotometer, Fourier Transform Infrared (FT-IR), X-Ray Diffraction (XRD), Field Emission Scanning Electron Microscopic (FE-SEM), and Transmission Electron Microscopy (TEM). The efficiency of the GNPs and conjugate to target and distribute in MCF-7 cells was evaluated using high fluorescent signals and confirmed by fluorescence microscopy. A variety of parameters were tested to investigate the anticancer potential of each compound toward MCF-7 cells in vitro. The conjugate demonstrated significant antioxidant effects with high tumor targeting efficacy and distribution in MCF-7 cells. It caused a decrease in cell proliferation rate and clonogenicity of MCF-7 cells, while initiating apoptosis and promoting cell cyceffects were coupled with upregulating the pro-apoptotic p53 while downregulating the anti-apoptotic Bcl-2, which resulted in an alleviation of the mitochondrial membrane potential (MMP) and, thereby, the secretion of cytochrome c. The outcomes of the present study propose the high feasibility for further development of the conjugate as a novel agent against cancer cells

    everal studies have been focused on identifying alternative medicines for the treatment of cancer. This study aimed to investigate the impact of B-Glucans extract (BG) against Ehrlich Ascites Carcinoma (EAC) induced toxicity and tissue damage in kidney. A total of 40 mice were assigned randomly to four groups (G1, control; G2, B-Glucans (75 mg/kg bw/day) orally for 2 weeks; G3, EAC and G4, EAC + BG). Current results revealed, EAC induced significant elevation in urea, creatinine, potassium (k+), chloride (Cl-) ions, kidney injury and significant decrease in sodium (Na+) and calcium (Ca++) ions when compared to B-glucans and control groups. Conversely, EAC + B-glucans modulate these changes and improved the renal functions and structure. EAC induced renal dysfunction and B-Glucans has the ability to improve renal functions. We can conclude that; BG could be used as an adjuvant therapy for kidney in ascites of hepatocellular carcinoma, Schistosoma and hepatitis

    The aims of this study were isolation-purification and characterization of l-glutaminase from L. gasseri BRLHM clinical isolates and investigation of its efficiency as an antimicrobial agent against multidrug-resistant P. aeruginosa. The MICs of l-glutaminase and gentamicin reference were evaluated by the well-diffusion method. The biofilm on the IUD contraceptive was visualized using atomic force microscopy (AFM) image analyses. The purified l-glutaminase possessed significant antimicrobial activity against P. aeruginosa isolates (p < 0.05), and the antibiofilm formation activity of the purified l-glutaminase was stronger than the antibiofilm activity of the referral standard drug, gentamicin (P < 0.05), which were checked by the inhibition of the biofilm formation on the IUD contraceptive device. Investigations indicated that l-glutaminase may have a crucial role in future clinical application

    Aims: To synthesize a silver-cored nanosuspension utilizing Ziziphus spina-christi fresh-leaf extract and evaluate their antimicrobial activity against multidrug-resistant pathogenic microbes. Materials and Methods: The prepared nanosuspension was analyzed by spectro-analytical techniques and tested for antimicrobial activity and resistance to biofilm formation. The leaf extract and nanosuspension were tested separately and together as a mixture. Results: Constituent nanoparticles were average-sized (∼34 nm) and were active against both Gram-positive and Gram-negative microbes and yeast. Candida albicans showed a 24.50 ± 1.50 mm inhibition zone, followed by Escherichia coli and Staphylococcus aureus. Increased bioactivity with the highest multifold increments, 150%, for erythromycin against all tested microbes was observed. Carbenicillin and trimethoprim showed 166%- and 300%-fold increments for antimicrobial activity against Pseudomonas aeruginosa, respectively. Conclusion: The nanosuspension exhibited strong potential as an antimicrobial agent and overcame multidrug resistance.

    Endometriosis is a chronic gynecologic disorder characterized by abnormal growth of endometrium-like tissues in the ectopic regions of the pelvic peritoneum. The pathophysiology of endometriosis is not completely understood; however, excessive endometrial cell proliferation together with resistance to apoptosis facilitates the migration, implantation, and survival of endometrial cells in the distant sites. Endoplasmic reticulum (ER) stress response (also called unfolded protein response) is a cellular defense mechanism triggered by ER stress. When severe enough, the so-called response initiates cell suicide, i.e., apoptosis. Therefore, therapeutic induction of ER stress in endometriotic cells could promote apoptosis and contribute to the management of disease. In this review, we discuss the pathogenic role of ER stress in endometriosis and the most recent findings regarding the induction of ER stress in connection with endometriosis.

    Gold nanoparticles (GNPs) act as less toxic and effective drug carriers with many specific properties that are particularly useful for biomedical and pharmaceutical applications. The present study was conducted to determine the anti-tumor properties GNPs. The new preparation technique is quick and cost-effective. GNPs formation was confirmed using various characterization techniques including X‐Ray diffraction (XRD) analysis, field emission scanning electron microscopy (FE-SEM), energy dispersive X-ray spectrometer (EDX), and transmission electron microscopy (TEM). XRD, FES-EM, and TEM experiments have confirmed the formation of GNPs which shows that the prepared particles are spherical inform and have a smooth surface with an average size of 24.32 ± 1.32 nm. Multiparametric analysis of GNPs toxicity at the level of individual cells using flow cytometry and cellular imaging-based approaches such as High Content Screening (HCS) have played key roles in the detection of toxicity and classification of compounds based on observed patterns of reversible and irreversible cellular injury. The effect of GNPs on the MCF-7 cell line regarding cell viability, cell nucleus morphology, membrane permeability, potential mitochondrial permeability, and cytochrome C release was further confirmed using a High Content Screening array scan via multi-parametric kit. Results indicated that the viable count of MCF-7 cells was highly decreased by treated with GNPs. Nuclei stained with Hoechst blue appeared to be mostly condensed in response to the treatment with the GNPs (1.5- fold increase in nuclear fluorescence intensity), with significant differences (p < 0.0263) as compared with untreated cells. Cell membrane permeability was significantly (p = 0.0019) upon GNPs showing an increase of 1.9-fold, as compared with the untreated MCF-7 cells. GNPs induce a significant (p = 0.0053) decrease in mitochondrial membrane potential by 32.7%, in comparison to the untreated cancer cells with significant induction of the release of cytochrome C (p = 0.0016) was recorded after exposure to the GNPs (0.43-fold increased mean intensity), The outcomes of the present study propose the high feasibility for using GNPs as a novel anticancer drug.

    Combining viruses and nanoparticles may be a way to successfully treat cancer and minimize adverse efects. The current work aimed to evaluate the efcacy of a specifc combination of gold nanoparticles (GNPs) and Newcastle disease virus (NDV) to enhance the antitumor efect of breast cancer in both in vitro and in vivo models. Two human breast cancer cell lines (MCF7 and AMJ13) and a normal epithelial cell line (HBL100) were used and treated with NDV and/or GNPs. The MTT assay was used to study the anticancer potentials of NDV and GNP. The colony formation assay and apoptosis markers were used to confrm the killing mechanisms of NDV and GNP against breast cancer cell lines. p53 and caspase9 expression tested by the qRTPCR technique. Our results showed that combination therapy had a signifcant killing efect against breast cancer cells. The fndings demonstrated that NDV and GNPs induced apoptosis in cancer cells by activating caspase9, the p53 protein, and other proteins related to apoptosis, which holds promise as a combination therapy for breast cancer.

The present study was conducted to determine the antioxidant and anti-tumor properties of fabricated GNPs-RGD (Arg-Gly-Asp peptide)-L-asparaginase (L-ASNase) conjugate. The preparation of GNPs-RGD-L-ASNase conjugate was firstly initiated by GNPs synthesis then encapsulation with SH-PEG-COOH accompanied by incubation with RGD and L-ASNase. The new preparation technique is quick and cost effective. GNPs and GNPs-RGD-L-ASNase conjugate formation were confirmed using various characterization techniques including: UV–visible spectroscopy, Fourier transform infrared spectroscopy (FTIR) analysis, X‐Ray diffraction (XRD) analysis, field emission scanning electron microscopy (FE-SEM), energy dispersive X-ray spectrometer (EDX) and transmission electron microscopy (TEM). UV-visible spectroscopy was used to detect the size and shape of the GNPs and GNPs-RGD-L-ASNase conjugate. The FTIR analysis was used to test the molecular states and functional groups of GNPs and GNPs-RGD-L-ASNase conjugate. XRD, FES-EM and TEM experiments have confirmed the formation of GNPs and GNPs-RGD-L-ASNase conjugate which shown that the prepared particles are spherical in form and have a smooth surface with an average size of 19.55±2.61 nm and 29.24±5.38 nm for GNPs and GNPs-RGD-L-ASNase conjugate respectively.     biocompatible peptides, although the effect of biocompatible GNP peptides on cellular responses is still not clearly understood. In the current study, GNPs with/without peptide were used as model probes to investigate the cytotoxicity to the human lung cancer cell line (A457) and human normal breast epithelial cell line (HBL-100). GNPs and GNPs-RGD preparation was confirmed and characterized using UV–VIS spectrophotometer, FE-SEM, FTIR, and TEM. The anticancer effect to A457 cell line was estimated using MTT assay. Our results show that the GNP-RGD had found significant tumor targeted efficacy and decrease in proliferation of A457 cell line compared with HBL-100 which appeared normal growth. Overall, our finding suggests a potential therapeutic effect of GNPs-RGD as a novel anti-cancer drug to be further developed

    H. pylori bacterium can directly detected from biopsy samples obtained from patient with gastroduodenal diseases using PCR. The study showed a various relationship between gastropathological cases and virulence genes ( ureA, cagA and vacA). Only vacA was not detected in gastric cancer biopsy and hence, it should be extensively focusing on the mechanism of action of the ureA, cagA genes in cancer diseases. Therefore, determination of H. pylori virulence genes may reveal information about regarding the risk and clinical outcomes in symptomatic patients with gastroduodenal diseases.

    Different plant sections from (Laurus nobilis, Malus domestica, and Citrus limon) were extracted using 80% methanol. All plants were then evaluated phytochemically for total flavonoids, with different concentrations of 952.77173.47, 980.55673.57, and 341.6683.33 mg/ml, respectively. Additionally, all plant extracts outperformed trolox in terms of their ability to reduce. Additionally, plants' radical scavenging abilities are stronger than vitamin C. (positive control). Some active plant chemicals, including hesperidin, quercetin-3-rhamnoside, and Q-3-O-B-glucopyranoside, underwent molecular docking. Due to its chemical structure, hesperidin had a stronger binding affinity with the active site of human kidenny type l-glutaminase (KGA) with a binding energy of (-7.09 kcal/mol) than the reference (crystal ligand), which had a binding energy of (-6.96 kcal/mol). Hesperidin peak is shown by HPLC analysis of Citrus limon methanolic extract at retention time 14.80 ppm. Purified hesperidin provided an inhibition of 65.33% for standard KGA in an in vitro inhibitory experiment when compared to CB-839 positive control. Using RT PCR, the GLS1 gene expression levels in the AMJ13 cell line were assessed after treatment with hesperidin and CB-839. The results revealed that hesperidin suppressed GLS1 gene expression in the cancer cell line while CB-839 increased GLS1 gene expression.
